扬州市蔷薇科植物PNRSV的鉴定与序列分析

Identification and Sequence Analysis of Prunus Necrotic Ring Spot Virus on Rosaceae Plants in Yangzhou,Jiangsu

  • 摘要: 为明确江苏省扬州市蔷薇科植物中李属坏死环斑病毒(Prunus necrotic ring spot virus,PNRSV)的发生情况,采用ELISA方法,以采集自扬州市不同地点的243份蔷薇科植物叶片样品为试材,测定其中4个分离物的外壳蛋白(CP)编码区序列,结合GenBank中已报道的相关序列对其进行多样性和系统发育分析。结果表明,江苏省扬州市蔷薇科植物中PNRSV的检出率为13%;序列分析表明本研究获得的PNRSV分离物与GenBank中其它286个分离物的核苷酸序列一致率为82% ~ 99%;系统发育分析表明PNRSV可以分成4个组,组间具有较为清晰的地理和寄主特异性,其中本研究获得的PNRSV分离物均位于PV32组。总之,PNRSV已经在扬州市发生并流行,应当引起重视。

     

    Abstract: In order to clarify the occurrence of Prunus necrotic ring spot virus (PNRSV) in Rosaceae plants in Yangzhou City,Jiangsu Province,the sequences of the coding region of the coat protein (CP) of four isolates were determined by ELISA on 243 leaf samples of Rosaceae plant collected from different spots in Yangzhou City,which were combined with the sequences of related sequences reported in GenBank for diversity and phylogenetic analyses. The results showed that the detection rate of PNRSV on Rosaceae plant in Yangzhou City,Jiangsu Province,China was 13%. Sequence analysis showed that the nucleotide sequences of the PNRSV isolates obtained in this study were in agreement with those of the other 286 isolates in GenBank at a rate ranging from 82% to 99%,and the phylogenetic analysis showed that the PNRSVs could be classified into four groups,with clearer geographic and host specificity among the groups. The PNRSV isolates obtained in this study were all located in the PV32 group. In conclusion,PNRSV has already occurred and become prevalent in Yangzhou City and should be taken seriously.
