Abstract:
Using the bulbs and scapes of the newly developed
Hippeastrum cultivar 'Xue Ran Chu Xia’ as explants, the optimal disinfection time for explants, and the best media for bud induction, proliferation, and rooting in tissue culture were investigated. The results showed that the optimal disinfection time with HgCl
2 was 20 minutes for both bulbs and scapes, with survival rates of 83.3% and 66.7%, respectively. The highest induction rate for bulbs reached 83.3% on MS medium supplemented with 2.0 mg·L
-1 6-BA, while the induction rate for scapes was 46.7% on MS medium supplemented with 1.0 mg·L
-1 6-BA. The optimal proliferation medium for adventitious buds was MS medium supplemented with 2.0 mg·L
-1 6-BA, 1.0 mg·L
-1 KT, 0.6 mg·L
-1 NAA and 30 g·L
-1 sucrose. The best rooting medium was 1/2 MS medium supplemented with 0.4 mg·L
-1 IBA, 15 g·L
-1 sucrose, and 0.2 g·L
-1 activated carbon, resulting in a rooting rate of 100%.
