The Tissue Culture Technology of Hippeastrum vittatum

  • 摘要: 以朱顶红新品种'雪染初霞’的鳞茎、花柄为外植体,筛选组织培养中最佳的外植体消毒时间,不定芽诱导、增殖和生根培养基。结果表明,鳞茎、花柄的HgCl2最佳消毒时间均为20 min,存活率分别达83.3%、66.7%。在MS+6-BA 2.0 mg·L-1培养基中,鳞茎诱导率高达83.3%;在MS+6-BA 1.0 mg·L-1培养基中,花柄诱导率为46.7%。不定芽的最适增殖培养基为MS+ 6-BA 2.0 mg·L-1+KT 1.0 mg·L-1 +NAA 0.6 mg·L-1 +蔗糖30 g·L-1;最适生根培养基为1/2 MS+IBA 0.4 mg·L-1+蔗糖15 g·L-1+活性炭0.2 g·L-1,生根率达100%。

     

    Abstract: Using the bulbs and scapes of the newly developed Hippeastrum cultivar 'Xue Ran Chu Xia’ as explants, the optimal disinfection time for explants, and the best media for bud induction, proliferation, and rooting in tissue culture were investigated. The results showed that the optimal disinfection time with HgCl2 was 20 minutes for both bulbs and scapes, with survival rates of 83.3% and 66.7%, respectively. The highest induction rate for bulbs reached 83.3% on MS medium supplemented with 2.0 mg·L-1 6-BA, while the induction rate for scapes was 46.7% on MS medium supplemented with 1.0 mg·L-1 6-BA. The optimal proliferation medium for adventitious buds was MS medium supplemented with 2.0 mg·L-1 6-BA, 1.0 mg·L-1 KT, 0.6 mg·L-1 NAA and 30 g·L-1 sucrose. The best rooting medium was 1/2 MS medium supplemented with 0.4 mg·L-1 IBA, 15 g·L-1 sucrose, and 0.2 g·L-1 activated carbon, resulting in a rooting rate of 100%.
